Prodrug reduces inflammation, restores physical function in aged mice

Experiments yielded promising results in efficacy, specificity, and non-toxicity

Prodrug (SSK1) found to effectively, selectively kill β-gal-positive senescent cells (SCs).

SSK1, first tested on mouse embryonic fibroblasts (MEFs) on healthy cells and counterparts.

To prove, gemcitabine-based chemical lacking β-gal-sensitive component was administered.

They used genetically modified cells which GLB1 gene was downregulated.

Gene knocked down SCs had regular levels of other markers, were unharmed by SSK1.

SSK1 was tested also for specificity for both murine and human cells.

It successfully eliminated SCs, causing little to no damage to healthy ones.

In induced lung fibrosis, it also succeeded in removing SCs from the affected lungs.

It proved superior when compared to three other senolytic candidate drugs.

Prodrug also successfully lowered number of activated macrophages aged mice livers.

Aged mice treated with SSK1 showed improved motor function, balance ability, among others.
